PPARγ激动剂对MKN-45胃癌细胞系增殖及MMP-7基因表达的影响

摘要:

目的:研究过氧化物酶增殖因子激活受体γ(peroxisome proliferators activated receptor γ,PPARγ)激活剂罗格列酮对人胃癌细胞系MKN-45增殖和MMP-7基因表达的影响.方法:不同浓度罗格列酮干预细胞后,用台盼蓝拒染法检测胃癌细胞株增殖活性,用RT-PCR法检测MMP-7基因表达情况.结果:罗格列酮作用24h明显降低MKN-45细胞的存活率,与时照组比较有显著差异(P<0.05),细胞存活率随着药物浓度增加而降低;MMP-7基因表达随着药物浓度增加而下降.结论:在胃癌细胞株MKN-45中,罗格列酮与PPARγ作用而激活下级信号传导,抑制胃癌细胞的增殖,且下调MMP-7基因的表达.

Objective To investigate the effect of PPARγ expressed in human gastric cancer cell MKN 45 on proliferation of gastric carcinoma cell line and MMP 7 gene expression. Method Reverse transcripition-polymerase chain reaction was used to demonstrate PPARγ and MMP 7 expression in MKN 45 cell line. Trypan blue was used to demonstrate the proliferation of cells. PPARγ agonist (rosiglitazone) reduced in a dose-dependent manner MMP 7 gene expression in the gastric cancer cells. Results Peroxisome proliferators activated receptor(PPAR)γ was expressed in MKN 45 cells. MKN 45 cells cultured by 10 μmol·L -1 rosiglitazone for 24 hours showed inhibitory effect and MMP 7 gene demonstrated a significant decrease. Conclusion These results suggest PPARγ agonist (rosiglitazone) can inhibit proliferation and expression of MMP 7 gene in gastric carcinoma cell line. And reduction of MMP 7 gene expression can be one of the mechanisms of the invasion and metastatic effect of PPARγ activation in human gastric cancer cells.
