一种新假定的解旋酶RuvBL1的抗凋亡活性研究

摘要:

目的:探索解旋酶RuvBL1与细胞凋亡之间的关系.方法:构建RuvBL1真核表达重组质粒,采用磷酸钙法转染L929细胞进行瞬时表达,通过转化生长因子(TNF)-α和放线菌素D诱导细胞凋亡,检测RuvBL1蛋白对凋亡的影响.结果:质粒构建后酶切得到约1.8kb长的片断,与RuvBL1的全长基因大小一致;转染L929细胞,可表达出相对分子质量为55000的蛋白,与理论值相同.RuvBL1组细胞凋亡率为40%,而对照组为70%;RuvBL1的抗凋亡活性呈剂量依赖性.结论:RuvBL1具有很好的抗凋亡活性,这一发现可能为凋亡相关疾病的治疗提供新的靶点.

Objective To explore the relationship between helicase and apoptosis. Method [WTBZ] The recombinant plasmid with full length RuvBL1 cDNA was constructed and expressed in mammalian cell by the standard calcium phosphate precipitation method. Result [WTBZ]Apoptosis assay using TNF α and 7 AAD co-stimulation showed that RuvBL1 was involved in anti-apoptosis activity. Conclusion This finding may be a new target for the therapy of apoptotic diseases.
