GDNF mRNA、GFR-α1 mRNA在不同年龄大鼠SVZ和海马表达的变化及其意义

摘要:

目的:观察不同年龄SD大鼠脑室管膜下区(SVZ)和海马GDNF mRNA及其受体GFR-α1 mRNA表达水平的变化,探讨其在神经发生及脑老化中的作用.方法:用RT-PCR方法检测胚胎18 d、新生以及1、3、8、12月龄SD大鼠SVZ和海马GDNF mRNA及其受体GFR-α1 mRNA的变化.结果:随着年龄增长,SD大鼠SVZ和海马GDNF mRNA及其受体GFR-α1 mRNA表达水平逐渐降低.且各年龄组海马表达参数的幅度小于SVZ.结论:GDNF及其受体GFR-α1在神经发育及脑老化进程中起到一定的作用.

Objective   To  investigate the effect of GDNF and GFRα_1 in  the subventricular zone and hippocampus on neural precursor proliferation,via analysing the gene expression of GDNFmRNA  and  GFRα_1mRNA from Sprague-Dawley rats of different ages. Methods   The level of GDNFmRNA and GFR-α_1mRNA in the subventricular zone and hippocampus  was observed and compared in rats of different age, using semi-quantitative RT-PCR method with β-actin as an inner consultation. Results   With the age increased,the gene expression of GDNFmRNA and GFRα_1mRNA in the subventricular zone and hippocampus was obviously decreased, and the gene expression of GDNFmRNA and GFRα_1mRNA in the subventricular outnumbered those of gene expression in hippocampus of different ages. Conclusions   The gene expression of GDNFmRNA and GFRα_1mRNA in the subventricular zone and hippocampus obviously decreases with the increase of age. GDNF and GFRα_1 exert important effects on the neural precursor proliferation, the decline of the neuronal compensatory ability and the neural plasticity of the aged brain.
