Objective A high performance liquid chromatography (HPLC) method was established for determination of melatonin in human serum with electrochemical detection. Methods The serum melatonin was extracted repeatedly by the chloroform,and was dried under vacuum in the organic phase. The residue was dissolved by the chloroform and dried under a nitrogen stream at room temperature. At last the residue was dissolved by the eluent for HPLC. Results The linearity of this method was up to 1.72-nmol·L -1; the detection limit was 0.084-nmol·L -1; the within-day imprecision was 4.1%~4.8%;the between-day imprecision was 4.4%~5.0%;the recovery was 76.5%~82.4%. Conclusion This method can be used for the determination of serum melatonin in clinical laboratories.
