高糖对肾小球系膜细胞c-fos表达与纤维结合蛋白分泌的影响

摘要:

目的:研究高糖对人肾小球系膜细胞纤维结合蛋白(FN)分泌以及c-fos表达的影响,探讨高糖对人肾小球系膜细胞的作用.方法:采用人肾小球系膜细胞进行体外培养,高糖作为刺激因素,ELISA法测定培养上清中FN含量,免疫细胞化学方法检测c-fos表达情况.结果:细胞培养上清中FN含量,在对照组每孔中为(88.9±19.1)-ng,高糖组(156.0±40.9)-ng,甘露醇组(156.0±21.9)-ng,后两组显著高于对照组(均P<0.05);c-fos的光密值,在对照组中为0.14±0.02,高糖组0.28±0.03,甘露醇组0.28±0.04,后两组显著高于对照组(均P<0.01).结论:高糖可增加人肾小球系膜细胞c-fos的表达,促进FN的分泌.

Objective  To investigate the production of fibronectin and expression of c-fos in mesangial cells cultured in high glucose and to explain the mechanism of diabetes nephropathy. Methods  The human mesangial cells were cultured in high glucose. The production of fibronectin was analysed by ELISA and the expression of c-fos was assaied by immunocellchemistry. Result  Compared with the contorol glucose,high glucose increased the expressoion of c-fos and the production of FN. Conclusion  The expression of c-fos and the production of FN increases when treated with high glucose.
