高糖对人肾小球系膜细胞TGF-β1 mRNA表达的影响及茶多酚对其干预作用

摘要:

目的:探讨高糖对人肾小球系膜细胞转化生长因子-β1(TGF-β1)mRNA表达的影响及茶多酚的干预作用.方法:运用RT-PCR法检测,TGF-β1在空白对照组、高糖组、茶多酚干预组和茶多酚对照组中0、12、36-h的表达情况.结果:高糖组TGF-β1-mRNA在3个时间点的吸光度值分别为0.330±0.014、0.549±0.027、0.761±0.032,与对照组相比有显著差异(均P<0.01),且36-h比12-h变化显著(P<0.01);茶多酚干预组分别为0.330±0.014、0.385±0.045、0.541±0.044,各时点均低于高糖组(均P<0.01),而与对照组无明显差异(均P>0.05).结论:高糖可导致人肾小球系膜细胞TGF-β1-mRNA表达的增加,而茶多酚可有效干预此效应.

Objective  To investigate the effect of high glucose on the expression of TGFβ  1mRNA in human mesangial cells and the intervention of tea polyphenols (TPs). Methods  To measure the expression of TGFβ  1mRNA by RTPCR in control group(C), high glucose group(G),high glucose+tea polyphenols group(G+T) and tea polyphenols control group(T) at  0, 12 and 36 hours respectively. Results  The expression of TGFβ  1mRNA increased in high glucose group. However, the expression of TGFβ  1mRNA was lower in TPs group compared with that in high glucose group. Conclusions  High glucose contributes to the increasing of TGFβ  1mRNA produced by human mesangial cells. TPs can reduce high glucoseinduced the expression of TGFβ  1mRNA.
