慢性GVHD狼疮样小鼠肾组织CTGF的异常表达

摘要:

目的:观察结缔组织生长因子(CTGF)在慢性移植物抗宿主病(cGVHD)狼疮样小鼠肾组织中的表达情况.方法:参照有关文献建立cGVHD狼疮样小鼠模型.按随机设计原则将模型小鼠分成模型组及正常对照组,每组6只.CTGF蛋白检测采用免疫组织化学及免疫荧光组织化学法,CTGF mRNA表达采用RT-PCR法检测;留取24 h尿,测定尿蛋白排泄率.结果:模型组小鼠24h尿蛋白总量明显高于正常对照组小鼠(P<0.01);RT-PCR检测到模型组小鼠肾组织CTGFmRNA表达较正常对照组小鼠明显升高(P<0.01);免疫组织化学及免疫荧光组织化学表明,正常对照组小鼠仅在少数肾小管有微弱CTGF表达,而模型组小鼠肾小管及间质均可见CTGF蛋白显著表达.结论:cGVHD狼疮样小鼠肾组织中CTGF蛋白及mRNA表达明显上调,可能在狼疮肾炎肾小球硬化及肾间质纤维化的进程中起重要作用.

Objective To investigate whether connective tissue growth factor(CTGF) modulates lupus nephritis(LN) in renal tissue of murine chronic graft versus host disease(GVHD) lupus nephritis.Methods Mice models of cGVHD lupus nephritis were establised according to relative literatures.The protein and mRNA expressions of CTGF were detected by immunohistochemistry,immunofluorescin histochemistry and RT-PCR in model group(n=6) and control group respectively.Proteinuria was also evaluated.Result The protein and mRNA expressions of CTGF were significantly increased in renal tissue of GVHD mice,respectively,which could be detected in tubular and interstitial cells.Conclusion CTGF may play a role in promoting glomerular and interstitial fibrosis in LN of murine chronic graft versus host disease.
