复方薤白胶囊对野百合碱诱导肺动脉高压大鼠ET、TNFα含量的影响

摘要:

目的:探讨复方薤白胶囊对野百合碱(MCT)诱导的肺动脉高压大鼠血浆中内皮素(ET)、肿瘤坏死因子α(TNFα)含量的影响.方法:SD大鼠60只,随机分为正常对照组、模型组、卡托普利组、复方薤白胶囊高剂量组和复方薤白胶囊低剂量组.采用MCT进行一次性腹腔注射(按60mg·kg-1)造模,22 d后测定平均肺动脉压、平均右心室压以及血浆中ET、TNFα的含量,并观察肺小动脉及右心室病理变化.结果:模型组平均肺动脉压、右心室压以及血浆中ET、TNFα含量明显高于正常对照组(P<0.01).其余各组平均肺动脉压、右心室压以及血浆中ET、TNFα含量明显低于模型组(P<0.05或P<0.01),且复方薤白胶囊高、低剂量组优于卡托普利组.结论:复方薤白胶囊能有效降低MCT诱导肺动脉高压大鼠的肺动脉压,可能的机制是抑制TNFα、ET产生,减轻MCT引起的炎性细胞浸润,改善肺小动脉及右心室重构.提示复方薤白胶囊将是防治肺动脉高压很有前途的中药,值得进一步研究.

Objective To explore the effects of compound Xiebai capsules(FFXBC) on endothelin(ET),tumor necrosis factor α(TNFα) content in blood plasma of rat model with pulmonary hypertension(PH) induced by monocrotaline(MCT).Methods Sprague-Dawley(SD) rats were randomly divided into five groups: normal control group,model group and three experimental groups in which the rats were fed with captopril,low dosage(single dosage) of FFXBC,high dosage(double dosage) of FFXBC respectively.The rat model was established by MCT intraperitoneal injection(60 mg·kg^{-1}),then the rats were treaded with gastric infusion respectively.Mean pulmonary artery pressure(mPAP),mean right ventricle pressure(mRVP),the ET and TNFα content in blood plasma were measured by radioimmunoassay method;histopathologic changes in lung including small pulmonary artery and right ventricle was observed.Results (22 days) after injection of MCT,the mPAP in rats of model group was significantly higher than that of normal control group(all P<0.01),TNFα and ET content in blood plasma were increased also(P<0.01),The mPAP,TNFα and ET content in rats of the rest groups were significantly lower than those of the model group(P<0.01 or P<(0.05)).Conclusion FFXBC is effective in reducing pulmonary artery hypertension,possibly through reducing the ET and TNFα producing,and improving small pulmonary arteries and right ventricles remodeling.It indicated that FFXBC is an effective compound of traditional chinese medicine,it will have a better prospect in preventing and treating COPD,PH and is worth further studying.
