Objective To observe the bio-effects on VEC-304 treated with low-frequency ultrasound and micro bubble agent,and to offer the clinic application theory basis of ultrasound.Methods Cells were divided into four groups :control(A);pure microbubble agent(B);pure ultrasound(C),according to the exposure time,group C was subdivided into C_1(exposure 60()s),C_2(exposure 90()s),C_3(exposure 120()s) and C_4(exposure 150()s);low-frequency ultrasound combined with micro bubble agent(D),according to the exposure time,group D was subdivided into D_1(exposure 60()s),D_2(exposure 90 s),D_3(exposure 120 s) and D_4(exposure 150()s).The survival rate of VEC-304 was detected by MTT colormetric assay.Visible spectrophotometric was applied to determine ROS and SOD.Apoptosis was analyzed by flow cytometry after ultrasonic exposure.Results The proliferation of group D were obviously inhibited,which was detected by MTT colormetric assay.The yield of ROS of groups C_3 and D_3 was higher than groups A and B,respectively.The yield of SOD of groups C_3 and D_3 was lower than groups A and B,respectively.The apoptosis rates were found to reach(11.57±2.68)% in group C_3 and(21.15±4.68)% in group D_3 by FCM.Conclusion Low-frequency ultrasound combinding with microbubble agent can induce VEC-304 injury and apoptosis,which is probably due to the adding yield of ROS in cultured liquid. |
