中药降氮汤对5/6肾切除大鼠肾小球结缔组织生长因子表达的影响

摘要:

目的:研究中药降氮汤对5/6肾切除肾小球硬化大鼠肾小球结缔组织生长因子(CTGF)表达的影响,探讨其防治肾小球硬化的作用机制.方法:采用5/6肾切除肾小球硬化大鼠模型,设立假手术组、模型组、氯沙坦组和中药降氮汤组.检测各组大鼠术后12周末血清尿素氮、肌酐及胆固醇水平.光镜及电镜观察第12周肾组织病理变化,并应用免疫组化法检测肾小球内CTGF的表达水平.结果:中药降氮汤组和氯沙坦组较模型组血清尿素氮、肌酐、胆固醇水平明显下降(P<0.01),两治疗组间比较无显著性差异(P>0.05).肾脏病理学检查显示,两治疗组肾小球增生硬化程度明显减轻.免疫组化显示两治疗组肾小球内CTGF表达较模型组明显减少(P<0.01),两治疗组比较无显著性差异(P>0.05).结论:中药降氮汤可显著抑制5/6肾切除肾小球硬化大鼠肾小球CTGF的过度表达,其防治肾小球硬化的作用机制与抑制CTGF的过度表达有关.

Objective To study the effect of Jiangdan Decoction on expression of connective tissue growth factor(CTGF) in glomeruli of 5/6 nephrectomized rats with glomerulosclerosis,and investigate its mechanism in preventing and treating glomerulosclerosis.Methods The 5/6 nephrectomized rats with glomerulosclerosis were assigned to the control group,model group,losartan treatment group and Jiangdan Decoction treatment group.The levels of blood urea nitrogen,serum creatinine and cholesterol were detected at the 12th weekend after operation.Renal pathology changes were observed at the 12th week by light microscope and electron microscope.Immunohischemistry was used to examine the expression of CTGF in glomeruli.Results The levels of blood urea nitrogen,serum creatinine and cholestrol in the Jiangdan Decoction group and losartan group were significantly lower than in the model group(P<0.01),every target was similar in the two treatment groups(P>0.05).The renal pathology examination showed that the degree of glomerular proliferation and glomerulosclerosis is significantly attenuated in the two treatment groups.Immunohistochemistry staining indicated that the expression of CTGF in glomeruli of the two treatment groups declined,respectively(P<(0.01)),this target was similar in the two groups(P>0.05).Conclusion Jiangdan Decoction can significantly inhibit the over-expression of CTGF in glomeruli of 5/6 nephrectomized rats with glomerulosclerosis,its mechanism in preventing and treating glomerulosclerosis is found relating to the inhibitory effect on the over-expression of CTGF.
