哮喘患者诱导痰上清液MIP-1α、MIF、MCP-1水平测定

摘要:

目的:通过测定哮喘患者诱导痰上清液肺泡巨噬细胞炎性蛋白-1α(MIP-1α)、肺泡巨噬细胞移动抑制因子(MIF)、单核细胞趋化蛋白-1(MCP-1)的水平,探讨三者在哮喘发病中的作用.方法:轻、中度哮喘患者14例,正常人12例,用高渗盐水进行痰诱导,痰液经解粘、离心后,对沉淀细胞分类计数,使用ELISA测定诱导痰上清液中MIP-lα、MIF、MCP-1浓度.结果:哮喘组诱导痰嗜酸粒细胞、淋巴细胞相对计数均高于正常组(分别为7.7%±1.4%、0.7%±0.8%,P<0.01;4.2%±1.0%、2.1±0.8%,P<0.01),而哮喘组诱导痰肺泡巨噬细胞相对计数构成比(57.8%±3.7%)明显低于正常组(65.9%±3.6%,P<0.01).哮喘患者诱导痰上清液MIP-1α质量浓度[(36.6±4.8)pg·ml-1]明显高于正常人[(32.6±1.7)pg·ml-1,P<0.05],两组诱导痰上清液MIF、MCP-1浓度无显著差异.哮喘患者诱导痰上清液MIF与嗜酸粒细胞相对计数呈正相关(r=0.53,P<0.05),与肺泡巨噬细胞、淋巴细胞相对计数无相关关系;哮喘患者诱导痰上清液中MIP-1α、MCP-1与肺泡巨噬细胞、嗜酸粒细胞和淋巴细胞相对计数均无相关关系.结论:MIP-1α可能参与哮喘的发病,嗜酸粒细胞可能通过释放MIF参与哮喘气道炎症.

Objective To determine the concentration of macrophage inflammatory protein-1 alpha(MIP-1α),macrophage migration inhibitory factor(MIF)and monocyte chemoattractant protein-1(MCP-1)in induced sputum supernate from asthmatac patients,and explore the roles of MIP-1α,MIF and MCP-1 in asthmatics.Methods Induced sputum samples were collected from 14 mild and moderate asthmatic patients and 12 healthy individuals.The differential counts of inflammatory cells were studied in induced sputum.The concentration of MIP-1α,MIF and MCP-1 in induced sputum supernate was measured by sandwich enzyme linked immunosorbent assay(ELISA).Results The percentages of eosinophils and lymphocytes in induced sputum supernate from asthmatic patients were higher than those from healthy individuals(P&lt;0.01).The percentage of macrophages from asthmatic patients was lower than that from healthy individuals((P&lt;0.05).)The concentration of MIP-1α in asthmatics was increased compared with that in healthy individuals((P&lt;0.05).)The concentration of MIF and MCP-1α had no difference between asthmatics and healthy individuals.The concentration of MIF in induced sputum supernate had close relationship to percentage of the eosinophils,but did not correlate to macrophages and lymphocytes.There was not any relationship between MIP-1α or MCP-1 and macrophages,eosinophils or lymphocytes.Conclusions The eosinophils and lymphocytes are important inflammatory cells in the pothgenesis of asthma.MIP-1α may play a role in asthmatics.Eosinophils may take part in the airway inflammation by producing MIF.
