B7-2分子在肝硬化患者肝脏组织中表达的Western blot检测方法研究

摘要:

目的: 建立肝硬化患者肝组织B7-2分子Western blot检测方法.方法:设置不同的一抗稀释比,筛选合适一抗稀释比.设置阳性对照、阴性对照,确认目的蛋白.结果:在一抗稀释比从1:50到1:200的范围内,蛋白显影灰度逐渐增加,随着一抗稀释比从1:200到1:1000的进一步递减,蛋白显影灰度却逐渐减弱.结论:在一抗稀释比为1:200条件下,B7-2分子在肝硬化患者肝脏组织中呈强阳性表达.

Objective To set up the method of Western blot for detection B7-2 protein in liver of patients with hepatic cirrhosis.Methods Western blot was used.The optimum dilution was determined by anlyzing protein grayscale in different B7-2 antibody dilutions.The B7-2 protein was confirmed by comparing with positive controls and negative controls.Result The developing grayscale of B7-2 protein in liver of human was increased in the range of 1∶50 to 1∶200,but decreased in the range of 1∶200 to 1∶1()000.Conclusion The B7-2 protein is strongly expressed in liver of the patients compared with the positive and negative controls under the condition of 1∶200 B7-2 antibody dilution.
