海藻糖对液态低温保存血小板保护作用的实验研究

摘要:

目的:研究海藻糖对液态低温(4℃)保存血小板的保护作用.方法:采集兔心脏血,按常规方法制备浓缩血小板(CP),在CP中加入不同浓度的海藻糖,4℃储存,用51Cr-铬酸钠进行标记后自体回输入兔体内,于回输后1、24、48、72 h抽取兔耳动脉血,测定放射性计数率,计算24、48、72 h血小板存活率,同时设常温对照组及单纯4℃储存的低温对照组进行比较.结果:加入不同浓度海藻糖各储存组的血小板寿命明显长于低温对照组,并且随着海藻糖浓度的增加而延长.结论:海藻糖可延长4℃液态保存血小板的寿命,对血小板具有保护作用.

Objective To study the protection of trehalose for liquid rabbit platelets stored at 4℃ traced by ^{51}Cr.Methods Cardiac blood was taken and made into concentrated platelet(CP).CP were stored at different conditions,such as low temperature and low temperature+different dose of trehalose for 24h.Platelets were transfused into rabbits’ bodies after being labelled with ^{51}Cr and platelet lifespan of 24,48,72h was calculated.Results After trehalose was added into the platelets stored at 4℃,the lifespan was obviously longer than that of platelets stored at low temperature.And with the dose increasing,the lifespan of platelets wes prolonged obviously till the dose was more than 50mg·ml^{-1}.Conclusion Trehalose,protecting platelets,can prolong the lifespan of liquid platelets stored at low temperature.
