Objective To choosing optimal test conditions with orthogonal experimental design method for determining cortisol and cortisone in urine by high-performance liquid chromatography.Method The key factors in the test were designed by orthogonal experimental method.According to the design the experiment was performed.The sample was extracted by solvent and analyzed by HPLC.Result A detailed analysis of parameters obtained showed that a satisfactory result may be obtained with only 36 tests as compared with 243(3~3+6~3)tests before.Conclusion This design is valuable in choosing conditions for determining trace materials by HPLC.
