大鼠三叉神经痛模型的建立

摘要:

目的:建立类似人类原发性三叉神经痛的大鼠模型.方法:雄性SD大鼠在YZ20-T6同光路手术显微镜直视下用铬肠线结扎眶下神经达到局部压迫的效果,术后两周行行为学观察并测量各组疼痛反应阈值,术后10周行病理形态学检查.结果:结扎眶下神经约两周后,大鼠对疼痛刺激反应的敏感性增加,疼痛反应阈值模型组与对照组比较有显著性差异(P<0.01);其病理表现类似人类三叉神经痛的神经病变脱髓鞘改变.结论:对大鼠三叉神经根慢性压迫性损伤可造成三叉神经痛样大鼠模型,其病理表现为三叉神经根的脱髓鞘改变,三叉神经半月节细胞未见明显异常改变.

Objective To set up a rat model of human trigeminal neuropathic pain.Methods The neuropathy was induced by a chronic constriction injury on the infraorbital branch of the trigeminal nerve(CCI-ION) under operative microscope.The changes of praxiology and leak point of pain irritant reaction were observed two weeks later.After ten weeks,the changes of pathomorphism were checked.Results The responsiveness of the CCI-ION rats to mechanical stimulation increased markedly on postoperative day of 12-14.Significant difference was observed between CCI-ION rats and contrasting group after analyzing the measured threshold value with statistic methods(P<0.01).Pathological changes in CCI-ION rats was like the demyelization in human beings’ trigeminal neuralgia.Conclusion The rat model of trigeminal neuropathic pain can be set up by a chronic constriction injury on the infraorbital branch of the trigeminal nerve.Demyelination is the main pathological changes in CCI-ION rats.No obvious change is observed in trigeminus ganglion cell.
