壬基酚对成年雌性SD大鼠生殖功能的影响

摘要:

目的:观察壬基酚对成年雌性SD大鼠生殖功能的影响.方法:对成年雌性SD大鼠每天灌胃壬基酚(50、100、200 mg·kg-1),阳性对照组每天灌胃乙蔗酚(0.2 mg·kg-1),阴性对照组管饲花生油.通过阴道涂片观察性周期的变化,28 d后处死大鼠,放免法测定血清雌激素、黄体生成素浓度,并作垂体、子宫、卵巢组织病理学检查,免疫组化技术检测子宫的雌激素受体(ER)表达情况.结果:壬基酚剂量≥100 mg·kg-1可使大鼠出现持续动情期,血清雌激素、黄体生成素水平降低(P<0.05),卵巢组织病理学切片显示卵泡数目减少(P<0.05),子宫ER表达无明显差异.结论:壬基酚在较高浓度时可抑制雌性大鼠的生殖功能.

Objective To investigate the effects of nonylphenol on the reproductive function of adult female SD rats.Methods Nonylphenol at concentrations of 50,100 and 200 mg·kg-1 and diethylstilbestrol at dose of 0.2 mg·kg-1 had been gavaged for 28 days.Rats were then sacrificed in the period of estrus for the examination of vaginal smears.Serum levels of 17β-estradiol(E2) and luteinizing hormone(LH) were measured by radioimmunoassay.Histopathological section of pituitary glands,uterus and ovaries were analyzed.By using SP immunohistochemistry,the estrogen receptorα(ERα) of the rat uterus were analyzed.Results The rats at doses of 100 mg·kg-1 and above appeared persitent estrus(PE).The serum E2 and LH levels were decreased with 100 mg·kg-1 or more(P<0.05).The histopathological section of ovaries showed the number of ovarian follicle treated with higher dose was decreased(P<0.05).The expressions of ER in uterus had no difference.Conclusion Nonylphenol at higher dose directly inhibites the reproductive function of adult female SD rats.
