氟达拉滨气道滴入对小鼠病毒性肺炎疗效观察

摘要:

目的:探讨氟达拉滨气道滴入对小鼠仙台病毒性肺炎是否具有治疗作用.方法:将21只ICR小鼠随机分为对照组、仙台病毒组和仙台病毒加氟达拉滨治疗组.接种病毒和用药7 d后评价各组一般状况、病理学表现,测定肺组织信号转导和转录活化因子-1(signal transduction and activation of transcription 1,STAT1)核酸和蛋白表达水平.结果:仙台病毒经鼻滴入可引起严重的肺部炎症,肺组织STAT1 mRNA和蛋白表达下降(P<0.05).氟达拉滨治疗组虽STAT1核酸和蛋白表达下降,但肺部炎症反应较仙台病毒组轻,淋巴细胞浸润程度较仙台病毒组轻(P<0.05),动物一般情况有所改善.结论:氟达拉滨气道滴入在仙台病毒导致的小鼠病毒性肺炎中具有减轻气道炎症、抑制淋巴细胞募集和改善动物一般状况的作用.该作用与氟达拉滨抑制STAT1合成的作用无关,而可能与其抑制淋巴细胞的作用有关.

Objective To investigate whether inhaled fludarabine has any therapeutic effects upon sendai virus infected mice.Methods 21 ICR mice were randomly divided into group A of controls,group B of mice infected with sendai virus and group C of mice infected with sendai virus and treated by inhaled fludarabine.7 days later,overall conditions,pulmonary pathological changes and signal transduction and activation of transcription 1(STAT1) expressions levels were evaluated.Results Sendai virus attack induced severe pulmonary inflammation and significant STAT1 declination both in protein and mRNA levels(P<0.05);fludarabine inhalation also induced significant STAT1 declination(P<0.05),but ameliorated sendai virus induced lung inflammation and improved overall conditions.Conclusions Fludarabine inhalation has therapeutic effects upon sendai virus infected mice.The mechanism may be explained by lymphocyte deactivation instead of STAT1 down-regulation.
