聚乙二醇修饰的胆固醇酯酶在高密度脂蛋白胆固醇均相测定中的作用

摘要:

目的:对胆固醇酯酶进行修饰,评价修饰酶的反应选择性.方法:采用各种聚乙二醇修饰剂对胆固醇酯酶进行修饰从而获得一系列修饰酶,然后研究这些修饰酶在高密度脂蛋白胆固醇的临床检验中的作用.结果:经双臂聚乙二醇修饰剂修饰的酶比用其他修饰剂修饰的酶具有更好的热稳定性和更高的酶活性,且表现出了较高的对高密度脂蛋白胆固醇的反应选择性.结论:表面活性剂对高密度脂蛋白的选择性解离作用非常重要,这有助于修饰酶发挥其对高密度脂蛋白胆固醇的反应选择性.

Objective To evalute reaction selectivity to high-density lipoprotein cholesterol(HDL-C) of modified cholesterd esterase.Methods Cholesterol esterase was modified with a series of polyethylene glycol(PEG) derivatives and investigated its potential clinical application for measurement of HDL-C.Result The enzyme modified by branched poly(ethylene glycol)derivatives exhibited greater retained activity,thermostability and selectivity to HDL-C than that treated with nonbranched PEGs.Conclusion The selective dissociating effect of the detergent on HDL is extremly important and helps the modified enzymes to selectively reacts to HDL-C.
