Midkine mRNA在食管鳞癌患者外周血中的表达及其临床意义

摘要:

目的:探讨靶基因中期因子(MK)在食管鳞癌患者外周血中的表达及其与食管鳞癌生物学行为的关系.方法:采用巢式RT-PCR方法检测54例食管鳞癌患者外周血的MK mRNA.10例正常健康志愿者外周血为阴性对照,11例食管鳞癌肿瘤组织为阳性对照.结果:54例食管鳞癌患者外周血MK mRNA阳性率为61.11%(33/54),并与食管鳞癌患者淋巴结转移有显著相关性(P<0.05),而健康成人外周血则无一例出现阳性.结论:应用巢式RT-PCR方法检测食管癌患者外周血中的MK mRNA具有较高的敏感性和特异性,它有望成为判断食管鳞癌恶性程度、转移、复发和监测疗效的客观指标.

Objective To investigate the expression of midkine in the peripheral blood of patients with esophageal squamous cell carcinoma(ESCC) and to analyze its relationship with clinicopathological features.Methods Nested RT-PCR was used to detect midkine mRNA in the peripheral blood from 54 patients with ESCC.The peripheral blood from 10 healthy volunteers and 10 specimens surgically removed out of ESCC were served as controls.Results The positive rate of midkine mRNA in 54 patients with ESCC was 61.11 %(33/54),and the mRNA expression of midkine was significantly correlated with lymph node metastasis of ESCC(P<0.05).But none of the peripheral blood samples was positive in 10 healthy volunteers.Conclusions The detection of midkine mRNA in peripheral blood from patients with ESCC using nested RT-PCR is highly sensitive and specific for the determination of ESCC.The method established may serve as a useful objective parameter for identifying the malignant behavior,metastasis,recurrence and therapeutic efficacy of ESCC.
