糖基化终产物对人肾小球系膜细胞结缔组织生长因子及纤维连接蛋白基因表达的影响

摘要:

目的:探讨体外培养条件下糖基化终产物(AGEs)对人肾小球系膜细胞结缔组织生长因子 (CTGF)及纤维连接蛋白(FN)基因表达的影响.方法:用不同浓度体外制备的AGEs干预系膜细胞及同一浓度的AGEs干预细胞不同时间.应用RT-PCR检测细胞中CTGF和FN的mRNA水平.结果:随着AGEs干预浓度的增加或干预时间的延长,系膜细胞CTGF和FN mRNA水平与对照组相比显著上升.结论:AGEs可导致人肾小球系膜细胞CTGF和FN mRNA的表达增加,在糖尿病肾病的发生、发展中起一定作用.

Objective To investigate the effects of advanced glycosylation end products(AGEs) on the expression of connective tissue growth factor(CTGF) and fibronectin(FN) in cultured human mesangial cells.Methods Human mesangial cells were cultured in DMEM medium and were incubated with AGEs modified bovine serum albumin(AGE-BSA).The mRNA of CTGF and FN were measured by reverse transcription polymerase chain reaction.Results The expression of CTGF mRNA and FN mRNA significantly enhanced by incubation of AGEs in a time-and dose-related manner(P<0.05).Conclusion The expression of CTGF and FN in human mesangial cells is upregulated by AGEs,which may have potential effects in the pathgenic mechanism of diabetic nephropathy.
