大鼠股骨闭合骨折模型的制作

摘要:

目的:研究大鼠标准股骨闭合骨折模型制作的方法.方法:简化设计并制作骨折造模支架.对78只雄性SD大鼠行逆行性克氏针固定股骨,然后使用造模支架制造股骨中断闭合骨折,通过X 线、组织学检查了解骨折类型、移位程度以及骨折的愈合情况.结果:术后X线摄片证实横断或短斜型骨折,移位不明显者67例,占总例数的85.9%,随访显示骨折呈典型的二期愈合过程.结论:简化改良自制造模支架制作的大鼠股骨闭合骨折成功率高,软组织损伤小,能最大限度减少外在因素对骨折的影响,通用性好,符合标准闭合骨折模型的要求.

Objective To develop a technique for the production of an experimental standard for closed fracture in rats.Method A reformed simplified fracture apparatus was designed and tested on 78 male Sprague-Dawley rats.First,the femur was treated with an intramedullary Kirschner’s wire retrogradely through a medial parapatellar approach.The femoral diaphysis was then fractured by means of a blunt guillotine driven by a dropping object.Results A highly reproducible transverse fracture with a success rate of 85.9% was produced.Minimal comminution and angulation of the intramedullary wires were found radiographically.After 12 weeks of follow up,a typical secondary fracture healing process was observed histologically.Conclusion The reformed apparatus is simple to build and easy to use.Through its application,a highly reproducible closed fracture model with minimal soft tissue damage is established.
