17β-雌二醇对小鼠淋巴结、脾脏和胸腺调节性T细胞及其Foxp3表达的影响

摘要:

目的:研究小鼠去卵巢及体内注射17β-雌二醇(E2)后,淋巴结、脾脏和胸腺CD4 +CD25 +调节性T细胞(regulatory T cell,Treg细胞)及相关调节性因子Foxp3表达的变化与E2的相关性.方法:采用流式细胞仪检测CD4+ CD25 +Treg细胞功能相关因子Foxp3的表达,CD4 +CD25 +T和CD4 +CD25+highT细胞数量的变化.结果:E2抑制淋巴结中Treg细胞Foxp3的表达,上调其在脾脏和胸腺中的表达;E2下调淋巴结中CD4 +CD25+ T细胞和CD4 +CD25+high T细胞的比例,上调它们在脾脏尤其是胸腺中的比例.结论:E2对淋巴结、脾脏和胸腺3种免疫器官中的Treg细胞有不同的调节作用.

Objective To investigate the effect of 17β-estradiol(E2) on CD4+ CD25+ Treg cells and the expression of their function relevance factor Foxp3 in the lymph nodes,spleen and thymus in vivo.Methods The expression of Foxp3 and the proportion of CD4+CD25+ T/CD4+ CD25+high T cells were detected by FCM.Results E2 could inhibit the expression of Foxp3 in lymph node,while upregulate it in spleen and thymus;E2 could downregulate the proportion of CD4+CD25+ T/CD4+ CD25+high T cells in lymph nodes,but upregulate them in spleen and thymus,especially in spleen.Conclusion E2 has different effect on Treg cells in lymph nodes,spleen and thymus.
