Objective To establish the orthotopic transplantation nude mouse model of pancreatic cancer for the study of its biological characteristics.Methods The subcutaneous tumors made by the subcutaneous injection of pancreatic cancer cell lines CFPAC-1 were used as the source of tissue for orthotopic implantation of tissue onto the pancreas,then the survival rates and spontaneous metastatic rate were compared with the two groups.Results There was no significant deviation in the rate of success,growth status and morphology between the two groups.The subcutaneous tumors were not metastasized,but the orthotopic implanted tumor metastatic rate,peritoneum metastatic rate,hepatic metastatic rate,pulmonary metastatic rate and lymph node metastatic rate were 87.5%,75.0%,37.5%,12.5% and 37.5% respectively.The orthotopic implanted tumor secreted CA19-9 as well.Conclusions The orthotopic implanted tumor model,more likely to mimic the process of tumor growth,local infiltration and metastasis,is superior to the subcutaneous implanted tumor model,thus it is ideal for the study of pancreatic cancer in vivo. |
