miRNA-21在激素非依赖前列腺癌细胞系PC3中的表达及对其增殖和侵袭性影响

摘要:

目的:探讨miRNA-21在激素非依赖前列腺癌细胞系PC3中的表达及对其细胞增殖和侵袭性的影响。方法:应用RT-PCR检查激素非依赖前列腺癌细胞系PC3在转染miRNA-21 抑制剂后miRNA-21的表达情况;用CCK-8和Transwell小室分别检测转染miRNA-21抑制剂后PC3细胞的增殖与侵袭能力。结果:RT-PCR显示,转染组miRNA-21的表达低于空白转染对照组(P<0.05);CCK-8和Transwell小室检测结果显示,转染组PC3细胞体外增殖及侵袭力明显降低(P<0.01)。结论:降低miRNA-21在激素非依赖前列腺癌细胞系PC3中的表达可抑制其增殖及侵袭力,这个发现为临床治疗非依赖性前列腺癌增殖及侵袭提供了新思路和方法。

Objective: To observe the effect of miRNA-21 on the proliferation and invasion of androgen-independent human prostate cancer cell line PC3. Methods: The difference of miRNA-21 expression between PC3-IN(miRNA-21 inhibitor) and PC3(control) cell lines was observed by RT-PCR; then the cell proliferation was examined by CCK-8 assay, and the invasive abilities of PC3 cell lines was observed by transwell invasion assay before and after treatment with miRNA-21 inhibitor. Results: The result of RT-PCR showed that miRNA-21 expression in PC3-IN cells was significantly lower than that in PC3 cell(P<0.05). Down-expression of miRNA-21 in PC3 cells could reduced the proliferation and invasion abilities of PC3 cells(P<0.01)significantly. Conclusion: Down-experssion of miRNA-21 plays an inhibitory role in the proliferation and invasion of PC3 cells.
