近红外影像评估脑淋巴瘤小鼠的靶向治疗

摘要:

目的: 通过7.0 T动物磁共振及近红外成像技术观察小鼠脑淋巴瘤模型,评估靶向药物的治疗情况。方法: 在立体定向仪下将人淋巴瘤细胞株Raji(CD20+)植入BALB/c裸小鼠大脑中,应用近红外成像技术评估荧光标记的靶向药物利妥昔单抗在小鼠体内分布,7.0 T动物磁共振及病理检查脑淋巴瘤,与近红外成像技术观察结果对照分析。结果: 通过近红外成像技术观察到,注射10 min后,荧光标记的利妥昔单抗在小鼠脑肿瘤实质部位逐渐聚集,持续3 h仍明显显像。磁共振显示肿瘤呈长T1长T2信号改变,边界不清,周围可见小片状水肿带,增强T1显示肿瘤实质部分明显强化;病理检查证实脑实质内肿瘤形成。结论: 立体定向种植法制作裸小鼠脑淋巴瘤模型简便易行,成瘤率高,近红外成像技术可以对荧光标记的靶向药物追踪,了解药物在体内的特异性分布。

Objective: To observe the brain lymphomas of the mouse model with the treatment of monoclonal antibody drug by 7.0T MR and near-infrared fluorescent (NIRF) screening. Methods: Human derived lymphoma cell line Raji(CD20+)was implanted into the right brain of BALB/c mouse by the stereotaxic technique. The distribution of the fluorescence labeled Rituximab in the mouse was revealed by near-infrared imaging. The results of the near-infrared imaging were compared with 7.0T MR and routine pathological examination. Results: Using near-infrared imaging, the fluorescence labeled Rituximab was detected to accumulation in the brain tumor of the mice just 10 minutes after injection, and lasted for 3 hours. The MR showed the tumor with higher signal in T1 and T2 weight image. The boundary of the tumor was unclear. The edema could be seen around the tumor. Enhanced T1 showed a substantial enhancement of tumor. The formation of the tumor was confirmed by pathological examination in accordant with the result of the near-infrared imaging. Conclusion: The brain lymphoma model of the mouse produced by the stereotaxic technique is simple and with high rate of tumor formation. Near-infrared imaging can track the fluorescence labeled targeted antibodies and show distribution in the body.
