锌-α2-糖蛋白对结直肠癌细胞株LoVo生物学行为的影响

摘要:

目的:初步探讨锌-α2-糖蛋白(ZAG)对结直肠癌细胞LoVo的生物学行为的影响。方法:构建和应用ZAG过表达质粒pGCMV/EGFP/Neo/ZAG转染结直肠癌细胞LoVo,免疫组化检测ZAG的表达变化,MTT法检测转染前后细胞增殖能力的变化以及细胞的克隆形成能力和侵袭力的改变。结果:过表达质粒pGCMV/EGFP/Neo/ZAG可成功诱导结直肠癌细胞LoVo表达ZAG,同时其细胞增殖力受到抑制、克隆形成率降低以及细胞的侵袭能力明显降低。结论:结直肠癌细胞LoVo的生物学行为的改变可能与ZAG的过表达相关,其在结直肠癌的发生发展过程发挥抑癌的生物学效应。

Objective: To explore the affection of adipokine zinc-α2-glycoprotein( ZAG) on the biological behavior in colorectal cancer LoVo cells. Methods: ZAG overexpression plasmid pGCMV/EGFP/Neo/ZAG was constructed and transfected into colorectal cancer LoVo cells, and the protein's expression of ZAG was detected by immunohistochemical. MTT assay was used to detect cell proliferation and the change of colony formation and invasive ability. Results: The overexpression of ZAG in colorectal cancer LoVo cells was induced, and the cell proliferation was significantly inhibited. Meanwhile, colony formation rate and cell invasion ability were significantly reduced comparing to the no-transfection-Lovo cells. Conclusion: The changes in the biological behavior of colorectal cancer LoVo cells may be correlated with the overexpression ZAG protein, which may have a tumor suppressor effect in the emergence and development of colorectal cancer.
