极低频磁场联合X线放疗对肝癌小鼠的治疗作用研究

摘要:

目的:探讨100Hz极低频磁场和X线放疗在肝癌小鼠治疗中的协同作用。方法:种植H22肝癌细胞建立肝癌小鼠模型,采用螺旋线圈制备磁通量0.7mT 的100Hz极低频磁场,直线加速器获得6MV的X线。根据治疗方式肝癌小鼠分为以下6组:对照组、单纯极低频磁场照射组、单纯4GyX线照射组、单纯8GyX线照射组、4GyX线联合极低频磁场照射组、8GyX线联合极低频磁场照射组,每组10只。在动物实验过程中每天记录肝癌小鼠的肿瘤直径以及生存天数。在联合4GyX线照射的基础上,进一步观察极低频磁场照射次数对肝癌小鼠肿瘤的抑制作用和生存时间的影响。不同组的生存时间比较采用Log-rank 检验,不同组间的肿瘤大小比较采用Student's T检验。结果:与对照组比较,单纯X线照射能显著缩小肿瘤直径,但同时缩短肝癌小鼠的总生存时间,并且单纯8GyX线照射组肝癌小鼠生存时间比单纯4GyX线照射组更短。与单纯极低频磁场照射组和单纯4GyX线照射组比较,4GyX线联合极低频磁场照射组肝癌小鼠的生存时间显著延长,并肿瘤直径显著缩小; 8GyX线联合极低频磁场照射组与单纯8GyX线照射组比较,肿瘤显著缩小且小鼠生存时间延长,但与单纯极低频磁场照射组比较,小鼠生存时间显著缩短。在联合4GyX线照射的基础上,更多次数的极低频磁场照射能明显改善肝癌小鼠的生存时间,缩小肿瘤体积。结论:极低频磁场与4GyX线在抗肝癌小鼠肿瘤生长和延长小鼠生存时间方面具协同效应。

Objective: To explore the possible synergism of 100 Hz extremely low frequency magnetic field(ELF-MF) and X-ray in treatment of hepatoma-implanted Balb/c mice. Methods: Hepatoma-implanted mice were established using H22 cells. Magnetic flux 0.7 mT to 100 Hz ELF-MF was preparaed by spiral coil and 6 MV X-ray irradiation was generated by linear accelerator. Hepatoma-implanted mice were divided into 6 groups:control group,group of ELF-MF alone,group of 4 Gy X-ray treatment alone,group of 8 Gy X-ray treatment alone,group of 4 Gy X-ray combined with ELF-MF,group of 8 Gy X-ray combined with ELF-MF,each with 10. Tumor growth and survival of H22 cells-implanted mice were evaluated by measuring the tumor diameter every day and recording survival days. Furthermore,tumor growth and mice survival were examined when different times of ELF-MF exposures combined with 4 Gy X-ray.Data referring to overall survival days and tumor diameters of the above groups were compared using Log-rank test and Student's T test. Results: Compared with control group,X-ray alone could significantly reduce the tumor size and shorten the survival days. Moreover,the survival in the group of 8 Gy X-ray alone was significantly shorter than that of 4 Gy X-ray alone. Compared with group of ELF-MF alone or group of 4 Gy X-ray alone,4 Gy X-ray combined with ELF-MF could significantly extend the overall survival days and reduce the tumor size. Compared with group of 8 Gy X-ray alone,8 Gy X-ray combined with ELF-MF could significantly extend the overall survival days and reduce the tumor size; but compared with group of ELF-MF alone,the survival days of mice were significantly shortened. And more times of ELF-MF exposure could further improve the survival and inhibit tumor growth in hepatoma-implanted mice when combined with 4 Gy X-ray. Conclusions: ELF-MF could possibly synergize with 4 Gy X-ray in terms of survival improvement and tumor inhibition of hepatoma-implanted mice.
