胸腺肽联合狂犬病疫苗接种对机体产生γ-干扰素、白细胞介素-2和中和抗体影响的研究

摘要:

目的:观察胸腺肽联合狂犬病疫苗接种对机体γ-干扰素、白细胞介素-2和中和抗体产生的影响。方法:Ⅱ级以下狂犬病暴露人群100例随机分为两组:疫苗组50例,单用狂犬疫苗2.5 U,常规免疫程序接种;胸腺肽组50例,给予50 mg单剂量肌肉注射+狂犬疫苗2.5 U,常规免疫程序接种。在注射前(D0)、注射7 d(D7)、注射6个月(M6)、注射12个月(M12)测定外周血淋巴细胞总数及γ-干扰素、白细胞介素-2水平,注射7 d、注射6个月、注射12个月测定中和抗体水平。结果:外周血淋巴细胞总数两组间无差异;胸腺肽组在注射后7 d白细胞介素-2水平和γ-干扰素水平均高于疫苗组(P<0.05);胸腺肽组在注射后7 d中和抗体产生量高于疫苗组(P<0.05)。结论:加用胸腺肽组注射后7 d中和抗体水平高于单用疫苗组,但6个月和12个月的抗体含量没有差异;两组接种后体内白细胞介素-2和γ-干扰素水平皆增高,白细胞介素-2增加的程度和抗体的产生量呈正相关。

Objective: To observe the effect of γ-interferon, interleukin-2 and neutralizing antibody levels in patients with rabies vaccination combination with thymosin. Methods: 100 patients were divided in to two groups. The vaccine group, 50 cases of rabies postexposure patients were given vaccine 2.5 U, routine immunization procedures; the thymosin group, 50 cases were given a single dose of thymosin 50 mg each with rabies vaccine 2.5 U together. Blood serums were maintained before injection(D0), on the 7 days(D7), on 6 month(M6) and 12 month(M12). γ-interferon, interleukin-2 and neutralizing antibody and lymphocyte count were detected together. Results: No lymphocyte counts differences was found in two groups. Leukocyte interleukin-2 and interferon-γ levels in thymosin group at 7 days after injection were higher than those in the vaccine group(P<0.05). Neutralizing antibody level was significantly higher in thymosin group than in vaccine groups on D7. Conclusion: The levels and positive rate of neutralizing antibodies in thymosin group are higher than those in vaccine group on D7, but there is no difference on M6 and M12. Interleukin-2 and γ-interferon levels are increased after inoculation vaccine, the level of interleukin-2 increased and the amount of antibody production is positively correlated.
