MTA1在胃癌细胞系和胃癌组织中的表达定位以及临床意义

摘要:

目的:探索肿瘤转移相关基因1( MTA1)在胃癌细胞系中的表达情况和定位,研究MTA1在胃癌组织中的表达和临床意义。方法:提取胃癌细胞系蛋白,使用Western blot 技术探测内源性MTA1的表达;选取高表达MTA1的胃癌细胞株,利用共聚焦激光扫描显微镜观察MTA1的定位;收集临床胃癌组织标本,测定MTA1的表达情况。结果:在8种胃癌细胞株中有6种胃癌细胞表达MTA1,主要定位在细胞核中。在胃癌组织中,高表达MTA1的比例达到63.6%(11对样品)。结论:胃癌细胞系中表达MTA1主要定位在细胞核中。胃癌组织中高表达MTA1,对胃癌临床诊断提供理论依据。

Objective: To explore the expression and the localization of MTA1 in the gastric cancer cell lines, and research the expression of MTA1 in the gastric cancer tissues and clinical significance. Methods: Total protein was extracted from gastric cancer cell lines. The expression of endogenous MTA1 was proved by Western blot. The localization of endogenous MTA1 in the gastric cancer cell lines was observed by using laser scanning confocal microscopy. The clinical gastric cancer tissues were collected, which were detected the expression of MTA1. Results: Six cell lines express MTA1 in eight gastric cancer cell lines, its localization was mainly in the nucleus. The rate of MTA1 overexpression was 63.6%(11 pairs) in the gastric cancer tissues. Conclusion: There are expression of MTA1 in the gastric cancer cell lines, which was expressed in nucleus. MTA1 was expressed in the gastric cancer tissues, it provide theoretical basis for clinical diagnosis.
