纳米雄黄脂质体的制备、特性检测和体外抗肿瘤细胞作用的研究

摘要:

目的: 研究纳米雄黄脂质体的制备方法并对其特性进行检测。方法: 采用化学沉淀法先制备纳米级雄黄,薄膜分散-高压均质法制备纳米雄黄脂质体。用电子显微镜、能谱仪、原子荧光光谱仪、激光粒度分析仪对其进行特性研究,并研究其体外抗肿瘤细胞的能力。结果: 薄膜分散-高压均质法制备的雄黄纳米脂质体平均粒径为102.3 nm,分散性良好,球形或卵球形。能谱仪证实其中含有砷和硫的成分,脂质体的药物包封率为82.78%。体外作用于HepG2肝癌细胞显示其有良好的抑制生长作用。结论: 采用上述方法可以成功制备纳米级别的雄黄脂质体,粒径较小,符合纳米制剂的要求,稳定性良好,药物包封率高,并且具备良好的抗肿瘤细胞的作用。纳米雄黄脂质体是一种新型纳米级剂型,为传统中药在抗肿瘤方面的应用提供了新的思路。

Objective To explore the preparation method and characteristics of realgar nano-liposome and its antitumor effect on liver cancer cells.Methods Realgar nanoparticles were prepared by chemical precipitation method and then realgar nano-liposomes were prepared by film dispersion and high pressure homogenizer method. TEM, EDS, ZETA-SIZER and atom fluorescence spectrophotometer were used to characterize realgar nano-liposomes. MTT assay was performed to examine the antitumor effect of realgar nano-liposomes at various concentrations.Results Results showed that the average diameter of realgar nano-liposomes was about 102.3 nm,and that the envelopment ratio of realgar was about 82.78%. The realgar nano-liposomes could produce an obvious cytotoxic effect on tumor cells in vitro.Conclusion Realgar nano-liposomes which can produce obvious antitumor effect on liver cancer cells can be successfully prepared by these methods. This study provides a new approach to the treatment of solid tumors by Traditional Chinese Medicine.
