应用组织芯片技术评估大肠癌手术切缘分子边界的初步研究

摘要:

目的: 通过分析p53、Ki- 67在癌组织和癌周不同距离(距离肿瘤1、2、3 cm)组织中的差异表达,探讨大肠癌手术切缘的分子边界。方法: 应用组织芯片技术和免疫组织化学方法研究44例大肠癌组织和癌周不同距离组织中p53、Ki- 67的表达情况。结果: p53、Ki- 67在大肠癌组织和癌周不同距离(距离肿瘤1、2、3 cm)组织中的表达呈现逐渐减弱的趋势,其递减表达差异具有统计学意义。结论: p53、Ki- 67所表现出的趋势性差异表达提示,在大肠癌手术切缘组织中存在分子边界。应用组织芯片大规模检测临床组织样本,具有快速、方便、经济、准确的特点。

Objective To investigate the surgical molecular margin through analyzing the differential expressions of p53 and Ki-67 in colorectal carcinoma and its surrounding tissue 1 cm,2 cm,3 cm away from lesion.Method Immunohistochemical assessment of p53 and Ki-67 expression was performed on a tissue microarray containing 44 cases of colorectal carcinoma and its surrounding tissue at different distance from lesion.Result The expressions of p53 and Ki-67 in colorectal carcinoma and its surrounding tissue at different distance indicated a tendency of decreasing gradually.The tendency of decreasing of p53 and Ki-67 expressions represented statistical significance.Conclusion The differential expressions of p53 and Ki-67 indicate the presence of the surgical molecular margin in colorectal carcinoma.Tissue microarray is rapid,convenient,economical and accurat in large-scale clinical application.
