低氧环境下转染血管内皮生长因子165对人胃癌细胞体外侵袭转移的影响

摘要:

目的 探讨低氧环境下血管内皮生长因子165(VEGF-165)对人胃癌细胞侵袭转移的影响及可能的机制。方法 利用腺病毒作为载体将已构建成功含VEGF-165基因的重组腺病毒(Ad-VEGF-165)瞬时转染人胃癌细胞株BGC-823,利用低氧诱导剂氯化钴进行低氧干预低氧转染(D)组,以单纯低氧干预(B)组及单纯转染(C)组作为对照,以未转染未行低氧干预的正常细胞作为空白对照(A)组,应用Transwell小室分析低氧环境下VEGF165转染前后胃癌细胞迁移能力的变化;利用半定量RT-PCR方法对4组细胞中uPA、MMP-2的mRNA表达水平进行分析。结果 D组胃癌细胞的侵袭能力及uPA、MMP-2的mRNA表达明显高于A、B、C组(P<0•05)。结论 低氧环境下转染VEGF-165能够明显提高人胃癌细胞BGC-823的侵袭能力及细胞中uPA、MMP-2的mRNA的表达,表明低氧环境下VEGF-165促进肿瘤细胞的侵袭转移。

Objective To evaluate the effects of VEGF165 on the invasion and metastasis of human gastric cancer cells line BGC-823 under hypoxia. Methods The BGC-823 cells were divided into 4 groups of transfected with Ad-VEGF165 transiently and induced by hypoxia mimic cobalt chloride (group D), induced by hypoxia mimic cobalt chloride alone (group B), transfected with Ad-VEGF165 alone(group C),and blank control(group A). A transwell cell culture chamber assay in vitro was used to detect changes in migration capacity of BGC-823 cells. The expressions of uPA and MMP-2 mRNA were detected by RT-PCR . Results The BGC-823 cells were transfected successfully. The mRNA expressions of uPA and MMP-2 in group D were significantly higher than those in groups of A, B and C (P< 0•05).Conclusion Hypoxia and transfection of VEGF165 into human gastric cancer cell line BGC-823 can significantly increase the expression of uPA, and MMP-2 indicating that VEGF165 under hypoxia promotes the invasion and metastasis of the tumor cells.
