乳腺癌组织中ADAM8 mRNA的表达及意义

摘要:

目的:探讨解整合素-金属蛋白酶8(ADAM8)mRNA在乳腺癌组织及正常乳腺组织中的表达量变化及其在乳腺癌发生、发展中的作用。方法:采用荧光定量逆转录聚合酶链反应(RT-PCR)方法检测39例乳腺癌及19例正常乳腺组织中ADAM8 mRNA 的表达,分析ADAM8 mRNA表达量与乳腺癌临床病理因素的关系。结果:39例乳腺癌组织中的ADAM8 mRNA表达水平低于19例正常乳腺组织(P=0.015);淋巴结转移组表达水平低于无转移组,Ⅱb+Ⅲ期表达水平低于Ⅰ+Ⅱa期,但表达差异均无统计学意义(P>0.05)。结论:ADAM8 mRNA在乳腺癌中表达下调,提示ADAM8基因表达抑制或缺失可能与乳腺癌的发生发展有关

Objective: To investigate the mRNA expression of ADAM8 in Breast cancer and the roles of ADAM8 in tumorigenesis and progression of Beast cancer.Methods:The ADAM8 mRNA expression in 39 breast cancer and 19 normal breast tissues was detected by flurence-quantitative RT-PCR, and the expression levels of ADAM8 were correlated with patients’clinical and pathological parameters. Results:Repressed ADAM8 mRNA expression was found in the 39 breast cancer tissues compared with that in 19 normal breast tissues(P=0.015).Furthermore, lower level of ADAM8 mRNA expression was found in stageⅡb、Ⅲ than in stage Ⅰ、Ⅱa,and the breast cancer tissues with positive lymph node metastasis exhibited less ADAM8 mRNA expression than those without lymph node metastasis, although this difference did not reach significant level (P>0.05). Conclusions:ADAM8 mRNA was lower expressed in breast cancer. Reduced or losed expression of ADAM8 gene may be involved in the pathogenesis and progress of this disease.
