Objective: To investigate the effect of Zebularine(Zeb), one of the demethylating agents, on the proliferation of human ovarian cancer cell line A2780 and its mechanism. Methods: A2780 cells cultured in RPMI1640 were treated with 50, 100, and 200μmol/L of Zeb. MTT assay, flow cytometry, and Annexin V-FITC staining were performed to analyze the cell proliferation activity, cell cycle, and apoptotic rate. Results: After A2780 cells were treated with Zeb of 100μmol/L and 200μmol/L for 24 to 72 hours, the cell proliferation activity was inhibited. The number of cells in G2 phase increased and the number of cells in S phase decreased significantly after the cells were treated with Zeb of 100μmol/L and 200μmol/L for 72 hours. The apoptotic rate increased significantly in cells treated with Zeb of 100μmol/L and 200μmol/L. Conclusion: Zeb could inhibit the growth of human ovarian cancer cell line A2780 ,which may due to block cell cycle and induce apoptosis. |
