zebularine对卵巢癌细胞的生长抑制作用

摘要:

目的 观察去甲基化制剂Zebularine对体外培养的卵巢癌细胞A2780的生长抑制作用,并进一步探讨其作用机制。方法 应用不同浓度的Zebularine (50μmol/L, 100μmol/L,200μmol/L)处理体外培养的卵巢癌细胞A2780,应用MTT法检测细胞增殖活性;流式细胞仪检测细胞周期分布; Annexin V-FITC染色法检测细胞凋亡率。结果 100μmol/L,200μmol/L的Zebularine处理卵巢癌细胞A2780 24~72h后,细胞增殖显著受到抑制(P<0.05);100μmol/L,200μmol/L的Zebularine处理72h后A2780细胞G2期细胞数量显著增加(P<0.05),S期细胞数显著降低(P<0.05);100μmol/L,200μmol/L浓度组细胞凋亡率均增加(P<0.05)。结论 Zebularine对卵巢癌细胞A2780具有生长抑制作用,其机制与阻滞细胞周期及诱导凋亡有关。

Objective: To investigate the effect of Zebularine(Zeb), one of the demethylating agents, on the proliferation of human ovarian cancer cell line A2780 and its mechanism. Methods: A2780 cells cultured in RPMI1640 were treated with 50, 100, and 200μmol/L of Zeb. MTT assay, flow cytometry, and Annexin V-FITC staining were performed to analyze the cell proliferation activity, cell cycle, and apoptotic rate. Results: After A2780 cells were treated with Zeb of 100μmol/L and 200μmol/L for 24 to 72 hours, the cell proliferation activity was inhibited. The number of cells in G2 phase increased and the number of cells in S phase decreased significantly after the cells were treated with Zeb of 100μmol/L and 200μmol/L for 72 hours. The apoptotic rate increased significantly in cells treated with Zeb of 100μmol/L and 200μmol/L. Conclusion: Zeb could inhibit the growth of human ovarian cancer cell line A2780 ,which may due to block cell cycle and induce apoptosis.
