H2-K/β2-m双转基因小鼠的制备及鉴定

摘 要: 在胎鼠原代神经元水平利用免疫荧光技术证实了H2-K在皮层神经元的表达,从C57 BL/6 小鼠脾脏中提取RNA,逆转录并扩PCR获得小鼠H2-K和β2-m基因并分别克隆入载体pBluescripte-Fzd9-Promoter中,然后双酶切获得Fzd9-Promoter-H2-K和Fzd9-Promoter-β2-m。将这两种基因同时显微注射至小鼠受精卵中制作双转基因小鼠,现已初步鉴定出三只双转的小鼠,和两只单转β2-m的转基因小鼠。

Abstract: Confirm the MHC class I molecules (H2-K and H2-Qa1) expression in the primary cultured mouse cortical neurons. In order to make double transgenic mouse over-expression functional MHC class I complexes specifically in the hippocampus, H2-K(flag) and β2-m were subcloned into plasmid pBlueScript-Frizzled9-promoter at the downstream of this hippocampal-specific promoter.. Three strains of H2-K/β2-m double transgenic mouses and two strains of β2-m transgenic mouses were generated now and ready for the further studying of MHC’s role in nerve system.
