全反式维甲酸对裸鼠子宫内膜癌治疗作用的实验研究

【摘要】 目的:研究通过建立人子宫内膜癌裸鼠模型,了解全反式维甲酸(ATRA)对裸鼠子宫内膜癌的作用及其机制。 方法:建立子宫内膜癌裸鼠模型,随机分为5组:较低剂量组(A组)、高剂量组(B组)、顺铂组(C组)、不同时间给予维甲酸组(D组)、空白对照组(E组)。治疗期间,测量皮下癌瘤大小并绘制生长曲线;观察抑瘤率;并用RT-PCR的方法检测癌组织中c-met RNA的表达。结果:建模成功后,高剂量组(B组40mg/kg)的抑瘤率(67.34%)较低剂量组(A组20mg/kg)(40.18%)高,而与顺铂组(C组68.06%)相当,B组与C组死亡率与也相当,两组相比无明显差异。各组裸鼠体重实验前后对比无显著性差异。裸鼠癌瘤中可检查测到肝细胞生长因子c-met的表达,维甲酸能有效抑制c-met的表达,与对照组及顺铂组相比差异有显著性,二组不同剂量与不同时间维甲酸组相比差异无显著性。结论:ATRA通过抑制c-met的表达抑制HGF/c-met介导的子宫内膜癌增殖,ATRA有希望成为治疗子宫内膜癌的药物之一。但实验中也发现ATRA并不能阻止移植瘤的形【摘要】 目的:研究通过建立人子宫内膜癌裸鼠模型,了解全反式维甲酸(ATRA)对裸鼠子宫内膜癌的作用及其机制。 方法:建立子宫内膜癌裸鼠模型,随机分为5组:较低剂量组(A组)、高剂量组(B组)、顺铂组(C组)、不同时间给予维甲酸组(D组)、空白对照组(E组)。治疗期间,测量皮下癌瘤大小并绘制生长曲线;观察抑瘤率;并用RT-PCR的方法检测癌组织中c-met RNA的表达。结果:建模成功后,高剂量组(B组40mg/kg)的抑瘤率(67.34%)较低剂量组(A组20mg/kg)(40.18%)高,而与顺铂组(C组68.06%)相当,B组与C组死亡率与也相当,两组相比无明显差异。各组裸鼠体重实验前后对比无显著性差异。裸鼠癌瘤中可检查测到肝细胞生长因子c-met的表达,维甲酸能有效抑制c-met的表达,与对照组及顺铂组相比差异有显著性,二组不同剂量与不同时间维甲酸组相比差异无显著性。结论:ATRA通过抑制c-met的表达抑制HGF/c-met介导的子宫内膜癌增殖,ATRA有希望成为治疗子宫内膜癌的药物之一。但实验中也发现ATRA并不能阻止移植瘤的形成,说明其有一定局限性,有必要采用联合用药以提高其抑瘤作用。

Abstract:Objective To investigate the effects of ATRA on established endometrial cancer bearing nude mice modle. Methods :Nude mice with transplanted HEC-1 tumor were randomly divided into five groups: middle-dose group(A group),high-dose group(B group), cisplatin group(C group),different time group(D group) ,blank control group (E group) . During the period of treatment,we measured the tumor size and drawed the growth curve of the tumors,The inhibitive rates of the tumor in nude mice was observed and detected the expression the c-met with the method of RT-PCR。Results: The inhibitive rate of the tumor was lower in middle-dose group than in high-dose group and cisplatin group (40.18% VS 67.34%,68.06%),but there is no difference in death rate of B group and C group,ATRA has inhibitive effect on c-met expression in tumor, the rate of c-met expression in tumor in group A、B and D is significant lower in group C and E. The c-met expression in tumor of group A and B group were similar to D group. Clusions:ATRA can inhibit the growth of tumor,which may be through the path of c-met by inhibiting the HGF/c-met.ATRA may be a potential drug to treat the endometrial cancer.but in our experimental study of ATRA,we discover the limitations of ATRA,It is necessary that ATRA comibined with other drugs can further inhibit the growth of tumor.
