壳聚糖修饰单壁碳纳米管的制备与细胞毒性研究

摘要:

目的:制备稳定的水溶性的壳聚糖/碳纳米管(CHI/SWCNT-COOHs)纳米材料以及对其表征,并检测纳米材料的细胞毒性。方法:市售单壁碳纳米管(Raw SWCNTs)以强酸处理, 形成残缺的羧基化单壁碳纳米管(SWCNT-COOHs),然后,低分子壳聚糖(CHI)与SWCNT-COOHs反应制备壳聚糖/碳纳米管(CHI/SWCNT-COOHs)纳米材料;运用透射电镜技术、拉曼光谱技术检测纳米材料的性状;通过台盼蓝染色计算纳米材料对A549细胞存活率的影响,以及计算纳米材料营养耗竭后对单细胞克隆的影响,运用扫描电镜检测对红细胞形态的影响。结果:CHI/SWCNT-COOHs在水系中很好的稳定性,对细胞无明显毒性,营养耗竭量减少,对红细胞形态没有显著影响。结论: CHI/SWCNT-COOHs性状稳定,具有纳米特性,细胞毒性低。

Objective: Used SWCNTs and CHI to synthesis CHI/SWCNT-COOHs as nano-materials, and tested it’s cells toxicity.Methods: Raw single walled carbon nanotubes (SWCNTs) were treated by acid oxidation to form Carboxylic SWCNT-COOHs. The amino of low molecular weight chitosan (CHI) and the carboxyl of SWCNT-COOHs formed nano-materials CHI / SWCNT-COOHs. The characters CHI/SWCNT-COOHs were determinated by transmission electron microscopy (TEM), Raman spectroscopy.A549 cells were treated with carbon nanotubes, and used Trypan blue cell viability assay test; Single-cell colonies statistics after CNTs nutrient depletion one week; Morphology Structural of the red blood cell by TEM with CNTs treated. Results: CHI/ SWCNT-COOHs with good stability in water systems improved the survival rate of cells, reduced nutrient depletion, and no significant effect on red blood cell morphology. Conclusion: CHI/SWCNT-COOHs had a stability in water, a nano-features, and low cytotoxicity.
