[Objective] To investigate the expression of HIST1H2AC and HIST1H2BC genes in pancreatic cancer cell lines Puta8988 treated with Pemetrexed, and verify the relationship between the genes and Pemetrexed-resistance. [Method] Pancreatic cancer cell lines Puta8988 were treated with two doses of Pemetrexed to get Puta8988+ and Puta8988++ with Pemetrexed-resistance, the expression levels of HIST1H2AC and HIST1H2BC and their genes in in Puta8988, Puta8988+ and Puta8988++ were detected by qRT-PCR and Western blot respectively. [Result] Results detected by qRT-PCR showed that expression levels of HIST1H2AC and HIST1H2BC genes were significantly up-regulated in Puta8988+ and Puta8988++ (P<0.05), for HIST1H2AC, up-regulated range was 4.64 times and 5.88 times of Puta8988 (control) respectively, for HIST1H2BC, 5.67 times and 23.07 times, respectively. Results detected by Western blot showed that expression levels of HIST1H2AC and HIST1H2BC were significantly up-regulated too in those cell lines with Pemetrexed-resistance (P<0.05), compared to control, the HIST1H2AC increased 54.35% and 87.43% in in Puta8988+ and Puta8988++, respectively, the HIST1H2BC did 189.26% and 774.85%, respectively. [Conclusion] That expressions of HIST1H2AC and HIST1H2BC and their genes in pancreatic cancer cell lines Puta8988 treated with Pemetrexed were significantly up-regulated, which implies that these genes possibly plays a role in Pemetrexed-resistance of pancreatic cancer cell lines.
