Objective: To investigate the clinical significance of non-coding RNA HOTAIR and MALAT1 expression levels in serum and bronchoalveolar lavage fluid (BALF) of patients with non-small cell lung cancer (NSCLC). Methods: Total RNA in serum and BALF was extracted from 86 NSCLC patients and 30 healthy people, and the expression levels of HOTAIR and MALAT1 genes were detected by real-time fluorescence quantitative RT-PCR, and their correlation with clinicopathological factors in NSCLC patients was analyzed. Meanwhile, the diagnostic efficacy of HOTAIR and MALAT1 in BALF was analyzed. Results: The expression level of HOTAIR in the serum of NSCLC patients was significantly higher than that of the healthy control group, but the expression level of MALAT1 was lower than that of the healthy control group, with statistically significant differences (P<0.05).The relative expression levels of HOTAIR and MALAT1 in BALF of NSCLC patients were significantly higher than those in the healthy control group. In addition, the relative expression level of MALAT1 in BALF of adenocarcinoma patients (1.03±0.26) was significantly higher than that of squamous cell carcinoma patients (0.64±0.08) (P<0.05).The sensitivity and specificity of HOTAIR in bronchoalveolar lavage fluid of NSCLC patients were 73.5% and 82.3%, respectively (cut off value was 5.16), while the sensitivity and specificity of MALAT1 were 85.4% and 87.6%, respectively (cut off value was 0.92). Conclusion: HOTAIR and MALAT1 are highly expressed in bronchoalveolar lavage fluid of NSCLC patients with high sensitivity and specificity, which has certain guiding value for the auxiliary diagnosis of NSCLC. |
