无创性延迟肢体缺血预适应对脑缺血再灌注大鼠存活素和血管内皮生长因子表达的影响

摘要:

目的:探讨无创性延迟肢体缺血预适应对脑缺血再灌注大鼠存活素和血管内皮生长因子(VEGF)表达的影响。方法:根据随机数字表法将90只大鼠分为假手术组(sham组)、脑缺血再灌注损伤组(I/R组)、脑缺血再灌注损伤+无创性延迟肢体缺血预适应组(I/R+NDLIP组),每组30只。测定各组大鼠神经缺损症状和行为评分、脑梗死范围,采用免疫组化二步法测定各组大鼠梗死灶周边皮质区存活素和VEGF阳性细胞,采用Western blotting法测定存活素和VEGF蛋白表达。结果:sham组大鼠未出现神经缺损症状,I/R+NDLIP组大鼠神经缺损症状和行为评分低于I/R组(P<0.05)。sham组大鼠未见脑梗死区域,I/R+NDLIP组大鼠脑梗死范围明显小于I/R组(P<0.05)。I/R组和I/R+NDLIP组大鼠梗死灶周边皮质区存活素和VEGF阳性细胞表达均高于sham组(P<0.05),I/R+NDLIP组大鼠梗死灶周边皮质区存活素和VEGF阳性细胞表达均高于I/R组(P<0.05)。I/R组和I/R+NDLIP组大鼠梗死灶周边皮质区存活素和VEGF蛋白相对表达量均高于sham组(P<0.05),I/R+NDLIP组大鼠梗死灶周边皮质区存活素和VEGF蛋白相对表达量均高于I/R组(P<0.05)。结论:无创性延迟肢体缺血预适应可降低脑缺血再灌注对神经的损伤,减小脑梗死范围,其机制可能与无创性延迟肢体缺血预适应可上调梗死灶周边皮质区存活素和VEGF的表达有关。

Objective:To investigate the effects of noninvasive delayed limb ischemic preconditioning on expression of survivin and vascular endothelial growth factor(VEGF) in rats with cerebral ischemia-reperfusion.Methods:90 rats were divided into sham group, cerebral ischemia-reperfusion injury group (I/R group), cerebral ischemia-reperfusion injury + noninvasive delayed limb ischemic preconditioning group(I/R+NDLIP group) according to random number method, 30 rats in each group. The neurological deficits and behavior scores, cerebral infarction were detected.Two-step immunohistochemical method was used to detect the survival rate and VEGF-positive cells in the cortex of the infarct.The expression of survivin and VEGF protein were detected by Western blotting. Results:No neurological deficits occured in rats of sham group, the scores of neurological deficits and behavioral in I/R+NDLIP group were lower than those in I/R group (P<0.05).The sham group had no cerebral infarction area,the cerebral infarction in I/R + NDLIP group was significantly smaller than that in I/R group (P<0.05).The survivin and VEGF positive cells in the cortex in the I/R group and the I/R + NDLIP group were significantly higher than those in the sham group (P<0.05).The survivin and VEGF positive cells in the cortex in the I/R + NDLIP group were higher than those in the I/R group (P<0.05).The relative expression of survivin and VEGF protein in the cortex of the I/R group and the I/R + NDLIP group were higher than those in the sham group (P<0.05).The relative expression of survivin and VEGF protein in the cortex of the infarct in the I/R + NDLIP group were significantly higher than those in the I/R group (P<0.05).Conclusion:Noninvasive delayed limb ischemic preconditioning can reduce cerebral ischemia and reperfusion injury to the nerve,reduce the scope of cerebral infarction,its mechanism may be related to noninvasive delayed limb ischemic preconditioning can upregulate the expression of survivin and VEGF in the cortex of the infarct.
