抗纤益心方通过TGF-β/Smads信号通路改善糖尿病心肌病大鼠心肌纤维化和心功能的研究

摘要:

目的:研究抗纤益心方是否通过转化生长因子(TGF)-β/Smads信号通路参与改善糖尿病心肌病(DC)大鼠心肌纤维化和心功能。方法:将40只大鼠随机分成4组各10只:对照组(C组)大鼠予单次腹腔注射生理盐水,抗纤益心方组(AF组)大鼠在C组基础上加用抗纤益心方溶液灌胃,DC组大鼠予单次腹腔注射链脲佐菌素制作DC模型,DC+AF组大鼠予单次腹腔注射链脲佐菌素和抗纤益心方溶液灌胃处理;观察4组大鼠经过不同处理后心功能及心肌纤维化变化情况。结果:DC组与DC+AF组大鼠血糖浓度明显高于C组(P<0.05);DC组大鼠左室舒张末压(LVEDP)显著上升(P<0.05),左室收缩压(LVSP)显著下降(P<0.05),DC+AF组大鼠LVSP明显高于DC组(P<0.05),LVEDP低于DC组(P<0.05);DC组与DC+AF组大鼠Ⅰ型胶原水平明显高于C组(P<0.05),DC+AF组大鼠Ⅰ型胶原水平显著低于DC组(P<0.05);DC组大鼠TGF-β1、Smad2及Smad3的磷酸化水平显著高于C组(P<0.05),而DC+AF组大鼠TGF-β1、Smad2及Smad3的磷酸化水平明显低于DC组(P<0.05)。结论:抗纤益心方对DC大鼠心肌纤维化具有一定的抑制作用,并能改善大鼠心功能,其作用机制与TGF-β/Smads信号系统密切相关。

Objective: To investigate the influence of Kangxian Yixin prescription on anti-cardiac fibrosis and improvement of cardiac function in diabetic cardiomyopathy rats through transforming growth factor β/smads signal pathway.Methods: Forty rats were randomly divided into 4 groups,i.e., control group(C group), anti-fibrinolytic group(AF group), diabetic cardiomyopathy group(DC group) and DC+AF group with 10 rats in each group. Rat models were induced by intraperitoneal injection of streptozotocin. After the models were established, the rats were treated with Kangxian Yixin prescription for 8 weeks by gavage. Rats in C group were given a single intraperitoneal injection of saline.In addition to a single intraperitoneal injection of saline, the rats in AF group were given Kangxian Yixin prescription by gavage. The rats were treated with streptozotocin by single intraperitoneal injection in DC group. Besides a single intraperitoneal injection of streptozotocin, the rats were additionally given Kangxian Yixin prescription by gavage in DC+AF group. The changes of cardiac function and cardiac fibrosis after different treatment were observed in the four groups.Results: The blood glucose concentration in DC group and DC+AF group was significantly higher than that in C group(P<0.05). The level of left ventricular end diastolic pressure(LVEDP) was significantly increased(P<0.05), and the level of left ventricular systolic pressure(LVSP) was significantly decreased(P<0.05). The LVSP in DC+AF group was higher than that in DC group(P<0.05). The LVEDP in DC+AF group was lower than that in DC group(P<0.05). The collagen-Ⅰ expression in DC group and DC+AF group was higher than that in C group(P<0.05), while the expression in DC group was higher than that in DC+AF group(P<0.05). The phosphorylation of TGF-β1, Smad2 and Smad3 in DC group was higher than that in C group(P<0.05) and DC+AF group(P<0.05).Conclusion: Kangxian Yixin prescription has certain effect on anti-cardiac fibrosis and improvement of cardiac function in diabetic cardiomyopathy rats, and its mechanism is closely related to the TGF-β/smads signal pathway.
