神经干细胞移植对脑缺血再灌注损伤大鼠脑脊液中炎症因子的影响

摘要:

目的:探索神经干细胞(NSCs)移植对大鼠脑缺血再灌注损伤后炎症反应的影响。方法:SD大鼠随机分为假手术组、磷酸盐缓冲液(PBS)对照组、NSCs移植实验组,对照组和实验组构建大脑中动脉闭塞再灌注(MCAO/R)模型,然后分别立体定位注射PBS和NSCs,在1、3、14、28 d对各组动物进行m-NSS评分和取脑脊液行ELISA检测。结果:(1)对照组、实验组大鼠神经功能都有恢复,而实验组更显著,m-NSS评分差异具有统计学意义(P<0.05)。(2)实验组脑脊液中肿瘤坏死因子α、白细胞介素1β含量在1、3、14 d都低于对照组,相对应的转化生长因子β1、白细胞介素10含量均高于对照组,差异均具有统计学意义(P<0.05)。结论:NSCs移植能抑制大鼠脑缺血再灌注损伤后炎症反应。

Objective: To investigate the effects of neural stem cells(NSCs) on inflammatory response in rats with cerebral ischemia-reperfusion model.Methods: Ischemia-reperfusion model of rats was built by middle cerebral artery occlusion and reperfusion(MCAO/R, occluded 90 min) method. Rats were randomly divided into sham operation group, phosphate buffer(PBS) control group, and NSCs transplantation group. Then on the 1st, 3rd, 14th and 28th days after stroke and transplantation, the neurological functions were assessed by m-NSS score, and the cerebrospinal fluid was collected and then detected by ELISA.Results: (1)The neurological function of NSCs transplantation group was significantly improved than that of PBS group with statistically significant difference(P<0.05).(2)The levels of TNF-α and IL-1β in cerebrospinal fluid of NSCs transplantation group were lower than those in the control group on the 1st, 3rd and 14th days, and the levels of TGF-β1 and IL-10 in NSCs group were higher than those in the control group, the difference being statistically significant(P<0.05).Conclusion: Transplantation of NSCs can alleviate the inflammatory response after cerebral ischemia-reperfusion injury in rats.
