葛根不同萃取部位对链脲佐菌素致2型糖尿病大鼠糖脂代谢影响的研究

摘要:

目的:通过葛根不同萃取部位对链脲佐菌素(STZ)诱导的2型糖尿病大鼠糖脂代谢影响的研究,探索葛根降血糖的主要有效部位。方法:SD雄性大鼠高脂高糖饲料喂养1个月后,腹腔注射STZ 30 mg·kg-1,建立2型糖尿病大鼠模型。将模型大鼠随机分为模型组、盐酸二甲双胍组(0.11 g·kg-1)及葛根的萃余部位组、乙酸乙酯部位组、二氯甲烷部位组和石油醚部位组,连续灌胃给药4周。于给药后每周测定大鼠空腹血糖(FBG)值,眼眶后静脉丛取血测定糖化血红蛋白(HbA1c)、空腹血清胰岛素(FINS)水平,计算稳态胰岛素抵抗(HOMA-IR)指数和胰岛敏感指数(ISI),并对血清中的甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)、游离脂肪酸(FFA)水平进行测定。结果:葛根二氯甲烷部位能明显降低糖尿病模型大鼠FBG、HbA1c、FINS、HOMA-IR指数、TG、TC、LDL-C、FFA,升高ISI。结论:葛根二氯甲烷部位、乙酸乙酯部位和萃余部位对2型糖尿病大鼠糖脂代谢具有较好的改善作用,其中以二氯甲烷部位作用最强。

Objective:To explore the effect of different extraction parts of Radix Puerarin on glucose and lipid metabolism in type 2 diabetic rats induced by STZ. Methods:SD male rats were fed with high fat and high sugar diet. After one month,the intraperitoneal injection of STZ 30 mg·kg-1 was conducted and type 2 diabetic rat model was established. The model rats were randomly divided into model group,metformin group(0.11 g·kg-1), Radix Puerariae raffinate site group,ethyl acetate group,dichloromethane fraction group and petroleum ether group,and the rats were continuously given intragastric administration for 4 weeks. The levels of fasting blood glucose(FBG), glycosylated hemoglobin(HbA1c)and fasting serum insulin(fins)were determined weekly and the steady-state insulin resistance(HOMA-IR)index and insulin sensitivity index(ISI)were calculated. The levels of serum triglyceride(TG),total cholesterol(TC),high density lipoprotein cholesterol(HDL-C), low density lipoprotein cholesterol(LDL-C)and free fatty acid(FFA)were also measured. Results:Dichloromethane fraction of Radix Puerarin significantly decreased the levels of FBG,HbA1c,FPG,fins,HOMA-IR,TG,TC,LDL-C and FFA,and increased the ISI. Conclusion:Dichloromethane and ethylacetate extract, and raffinate parts of Puerarin can improve glucose and lipid metabolism in type 2 diabetic rats,of which the dichloromethane extract shows the best effect.
