Objective:To explore the effect of different extraction parts of Radix Puerarin on glucose and lipid metabolism in type 2 diabetic rats induced by STZ. Methods:SD male rats were fed with high fat and high sugar diet. After one month,the intraperitoneal injection of STZ 30 mg·kg-1 was conducted and type 2 diabetic rat model was established. The model rats were randomly divided into model group,metformin group(0.11 g·kg-1), Radix Puerariae raffinate site group,ethyl acetate group,dichloromethane fraction group and petroleum ether group,and the rats were continuously given intragastric administration for 4 weeks. The levels of fasting blood glucose(FBG), glycosylated hemoglobin(HbA1c)and fasting serum insulin(fins)were determined weekly and the steady-state insulin resistance(HOMA-IR)index and insulin sensitivity index(ISI)were calculated. The levels of serum triglyceride(TG),total cholesterol(TC),high density lipoprotein cholesterol(HDL-C), low density lipoprotein cholesterol(LDL-C)and free fatty acid(FFA)were also measured. Results:Dichloromethane fraction of Radix Puerarin significantly decreased the levels of FBG,HbA1c,FPG,fins,HOMA-IR,TG,TC,LDL-C and FFA,and increased the ISI. Conclusion:Dichloromethane and ethylacetate extract, and raffinate parts of Puerarin can improve glucose and lipid metabolism in type 2 diabetic rats,of which the dichloromethane extract shows the best effect. |
