Objective To investigate the effect of high glucose on reactive oxygen species (ROS) in human mesangial cells and the intervention of tea polyphenols(TPs).Method Colorimetric assay was used to detect SOD、GSH?PX、MDA、O 2 -· in the supernatant of high glucose culture of human mesangial cells treated with or without TPs.Results SOD and GSH?PX were lowered,while MDA and O 2 -· increased in the supernatant of high glucose culture without TPs.However,SOD 〔( 32.507± 3.368)NU·ml -1 〕 increased with TPs compared with that only with high glucose,GSH?PX〔(56.233±2.808) NU· ml -1 〕increased as well,while MDA〔(3.280±0.149)μmol·L -1 〕and O -· 2〔(29.751±2.528)N·L -1 〕decreased.Conclusion High glucose contributes to an increase in ROS produced by human mesangial cells and TPs can better control high glucose induced oxidative stress. |
