糖基化终产物对人肾系膜细胞MMP-2表达的影响

摘要:

目的:观察糖基化终产物(advanced glycosylation end products,AGEs)对人肾系膜细胞(human renal mesangial cell,HRMC)基质金属蛋白酶-2(matrix metalloproteinase,MMP-2)表达的影响.方法:运用糖基化修饰的牛血清白蛋白(AGE-BSA)干预体外培养的HRMC,Western blot检测系膜细胞MMP-2蛋白表达.结果:HRMC的MMP-2蛋白表达水平随AGE-BSA浓度增加和干预时间延长而降低(P<0.01).结论:MMP-2可能参与AGEs所致的糖尿病肾病的发生发展.

Objective To observe the effects of advanced glycosylation end products(AGEs) on the expression of matrix metalloproteinase-2(MMP-2) in cultured human renal mesangial cells(HRMC).Methods HRMC were incubated with different concentrations of AGE-BSA for different time.The protein expression of MMP-2 in HRMC were analyzed by Western blot.Result AGE-BSA decreased the expression of MMP-2 in HRMC time and dose dependently((P<0.01)).Conclusion The data indicates that MMP-2 might mediate the role of AGEs in the development of diabetic nephropathy.
