糖尿病大鼠肾脏细胞外基质金属蛋白酶诱导因子和基质金属蛋白酶2的表达

摘要:

目的:研究糖尿病大鼠肾脏组织中细胞外基质金属蛋白酶诱导因子(EMMPRIN)和基质金属蛋白酶2(MMP-2)的表达,探讨糖尿病肾病的发病机制.方法:将SD大鼠随机分成正常对照组(NC组)和糖尿病模型组(DM组),DM组腹腔注射链脲佐菌素(55 mg·kg-1)诱导糖尿病大鼠模型,于第4、第8周分批处死大鼠,用免疫组化检测EMMPRIN、MMP-2表达,图像分析仪分析.结果:正常大鼠EMMPRIN、MMP-2主要表达在肾小球系膜细胞、内皮细胞、足细胞、球囊壁层上皮细胞、肾小管上皮细胞,造模后4、8周时DM组大鼠EMMPRIN、MMP-2表达较正常对照组明显下降.结论:糖尿病时肾小球EMMPRIN和MMP-2表达水平的降低,可能是引起细胞外基质积聚的原因之一.

Objective To investigate the expression of extracellular matrix metalloproteinase inducer(EMMPRIN) and matrix metalloproteinase-2(MMP-2) in diabetic rat’s kidneys.Methods Rat diabetic model was induced by streptozotocin(55mg·kg^{-1}),and kidneys were examined pathologically and the expression of EMMPRIN and MMP-2 was studied by immunohistochemistry.The intensity of EMMPRIN and MMP-2 was analyzed by imaging quantitative analysis technique.Results EMMPRIN and MMP2 were mainly expressed in the mesangial cells,endothelial cells,podocytes,parietal epithelial cells of Bowman’s capsule,and tubular cells.The expression of EMMPRIN and MMP-2 was significantly weakened in the glomeruli of diabetic rats than that of the control animals(P<0.05).Conclusion Expression of EMMPRIN and MMP-2 in glomeruli is markedly decreased in diabetic rats,which is involved in the development of diabetic nephropathy.
