Objective To investigate the possibility of the mixed culture of detergent-extracted muscle scaffold originated from autologous muscle seeded with autologous Schwann cells used to be nervous graft. Methods Schwann cells were cultured and proliferated in vitro.Detergent-extracted muscle scaffold was prepared by using chemical extraction and pretreated by polymer of lynines, then they were mixedly cultured for two weeks.Their growth and adhesion were observed by scaning electron microscope.Result Schwann cells adhered well to the autologous detergent-extracted muscle scaffold, and then proliferated and migrated on the scaffold, finally formed a Schwann cells columns just like Büngner’s band. Conclusion The artificial nerve model of autologous detergent-extracted muscle scaffold seeded with Schwann cells in vitro is suitable to repair nerve defect. |
