新生鼠脑缺氧缺血性损伤后海马区细胞增殖的观察

摘要:

目的:观察新生大鼠缺氧缺血性脑损伤(HIBD)后海马区细胞增殖的情况,探讨脑组织内源性修复的可能机制.方法:建立新生大鼠HIBD模型,分假手术组、单纯缺氧组、缺氧缺血组.于脑损伤后不同时间点取脑,分别行HE染色和BrdU免疫组化检测.结果:缺氧缺血组于缺氧缺血后3d BrdU阳性细胞有表达,7~14d达到高峰,21d时减少,在各时间点均高于其他组(P<0.05).结论:HIBD后海马区存在细胞增殖,推论新生大鼠在出生后早期能显著抵抗HIBD,可能产生对海马的保护作用.

Objective To observe the expression of proliferation in hippocampus following hypoxic-ischemic brain damage(HIBD) in neonatal rats and to explore the possible endogenous neurogenesis of brain.Methods Neonatal HIBD rat model was established and the rats were divided into sham group,hypoxia group,hypoxia-ischemia(HI) group.Brain of rats was obtained at different time points after HIBD to conduct HE staining,immunohistochemistry of BrdU.Results In the HI group,the level of BrdU-positive cell increased on the 3rd day of HIBD,reaching the peak on the 7th-14thday,and decreasing on the 21stday,all higher than that in other groups at different time(all P<0.05).Conclusion There is proliferation in hippocampus after HIBD.It is suggested that neonatal rat hippocampus is remarkably resistant to HI damadge,which possibly generates the protection for the hippocampus.
