糖基化终产物对大鼠血管平滑肌细胞骨保护素mRNA表达的影响

摘要:

目的:研究糖基化终产物(AGEs)对大鼠血管平滑肌细胞(VSMCs)骨保护素(OPG)mRNA表达的影响.方法:体外培养VSMCs,在含10mmol·L-1β-甘油磷酸(β-GP)的培养液中加入不同浓度(0、50、100、200、400mg·L-1)的AGE-牛血清白蛋白(BSA)与VSMCs 孵育不同时间(0、12、24、36、48、72h).采用RT-PCR检测OPG的mRNA表达水平.结果:AGE-BSA呈时间和剂量依赖性地增加VSMCsOPGmRNA的表达;200mg·L-1的AGE-BSA与VSMCs孵育24h,OPGmRNA表达水平达到高峰.结论:AGE-BSA能够促进大鼠VSMCsOPGmRNA的表达.

Objective To investigate the effect of advanced glycation end products(AGEs) on the expression of osteoprotegerin mRNA in cultured rat vascular smooth muscle cells(VSMCs).Methods VSMCs were cultured in vitro for 3~8 generations,and then exposed to β-glycerophosphate or β-glycerophosphate plus AGE-BSA of different concentrations for different time duration.OPG mRNA was detected by semi-quantity RT-PCR.Results OPG mRNA levels were greatly increased in a concentration and time-dependent manner as compared with the group without AGE-BSA treatment(P<0.05).OPG mRNA expression increased mostly in a concentration of 200mg·L-1 and at the time of 24h.Conclusion AGE-BSA potentially upregulates the expression of OPGmRNA to a certain extent in rat vascular smooth muscle cells.
