Abstract: Objective: By producing the rat models with endotoxic shock, to detect serum TNF-α, corticosteroid , LPS level and adrenal glands tissue LPS level,and to observe histological changes of adrenal gland tissue with microscopy. Methods: Forty Wistar rats were randomly divided into two groups: normal control group (n=10), endotoxic shock group (n=30). Endotoxic shock group were sensitized by D-GalN by intraperitoneal injection, then given LPS by tail vein injection. Changes of mean arterial pressure (MAP) were observed to evaluate the animal model. Serum TNF-αconcentration, serum and adrenal glands tissue LPS concentrations were measured by enzyme-linked immunosorbent assay(ELISA). Serum corticosteroid concentration was detected by Electro-Chemiluminescence Immunoassay(ECLI). Histological alterations of adrenalglands tissue were studied with microscopy. Results: The MAP decreased obviously after injection of LPS, then recovered to normal, and decreased slowly, and kept at a low level. 19 rats corresponded shock standards, whose serum TNF-α, corticosteroid and LPS concentrations were significantly higher than those of the normal control group(p<0.01), and adrenal glands tissue LPS concentration was also significantly higher than that of the normal control group(p<0.05). Pathological changes of adrenal glands tissue presented congestion and dropsy. Conclusion: LPS、TNF-αinvolves genesis and development of endotoxic shock. Serum corticosteroid of endotoxic shock rats increases obviously, and the adrenal glands tissue presented pathological changes. |
